Principle
Signosis' TF Gaussia Luciferase Reporter Stable Cell Lines utilizes a specially engineered promotor construct to express Gaussia Luciferase under the activation of your desired TF of interest. The promoter region consists of multiple repeats of a cis-element TF binding site, a DNA fragment from the promoter region of a known TF downstream gene, or a DNA fragment containing putative/known TF binding sites. Once activated, the TF translocates to the nucleus and often interacts with relevant co-factors to drive Gaussia Luciferase gene expression.
The Gaussia Luciferase is a gene from the copepod Gaussia Princeps. This special luciferase is a small, naturally secreted luciferase of approximately 20kDa that is significantly brigher than traditional luciferases. Because this luciferase is naturally secreted from the cells, the lysis step can be skipped, simplifying and streamlining your experimental procedures. Therefore, a reporter system utilizing this allows for an easy, direct, noninvasive, and continuous measurement of reporter activity from the same population of cells.
Gaussia Luciferase requires a coelenterazine-based luciferase substrate to produce light. At Signosis, we provide our own one-step Gaussia Luciferase Substrate that is easy to use and highly sensitive, which can be purchased along with your cell line of choice.
