The pMSCV-EGFR wt vector contains the wt EGFR gene under the MSCV promoter for high expression in hematopoietic, HEK293, and stem cells. Downstream of the EGFR gene is a DYKDDDDK-tag for Western blotting. Furthermore, the coGFP under the EF1 promoter can be used as a selection marker with stable cell lines. The base vector is a modified pMSCV vector with the LTRs and the lentiviral packaging signal (?) removed for standard expression. Epidermal growth factor receptor (EGFR) is a transmembrane tyrosine kinase receptor that plays a critical role in regulating cell division and death. The EGFR gene is present on chromosome 7p11.2 and has 28 coding exons in which exons 5�7 and 13�16 code for the ligand binding domain and exons 18�24 code for the TK domain.  Downstream effects of EGFR activation include modulation of three major pathways of RAS-RAF-MAPK, PI3K/AKT and JAK/STAT pathways which can lead to cell survival, proliferation and migration of cancer cells.
