CAR EMSA (Electrophoretic-Mobility Shift Assay) kit is used for monitoring CAR (constitutive androstane receptor) DNA binding activities and activation. CAR is a member of the nuclear receptor superfamily but it can act as a transcription factor. Ligand binding results in translocation of CAR from the cytosol into the nucleus, where the protein can bind to specific DNA sites, called response elements resulting in activation or repression of target gene transcription. Upon activation, CAR can recognize and interact with the unique DNA binding sequence and form protein/DNA complex, which can be separated from free DNA due to the differences in their electrophoretic mobility in native (non-denaturing) polyacrylamide gels.
