Principle
When a transcription factor (TF) is activated, it binds to a biotin-labeled probe designed based on its consensus binding sequence. This TF/probe complex, when run through electrophoresis, migrates slower than the free probe, resulting in a distinguishable shifted band. By comparing the density of these shifted bands across different samples, variations in TF activation levels can be easily determined.
